Several recent studies have demonstrated that expression of human Xbox binding protein 1 ( XBP1) is associated with ERstatus in breast tu mors and overexpressed in a subset of breast tumors. XBP1 has two splicing variants, which were designated as XBP1S and XBP 1U, respectivel . However, little is known about the expression pattern of XBP1S and XBP1U in breast cancer cells and about their roles in ERsignaling. In this stud , the expression of two splicing forms of XBP1 was detected in breast cancer cell lines with RTPCR. Estrogen response element ( ERE)containing luciferase reporter assa was used to determine the effects of XBP1S and XBP1U on the transcription activit of ER in MDAMB435 breast cancer cells. The result showed that both XBP1S and XBP1U enhanced the transcription activit of ER in a hormoneindependent and dosedependent manner and the activit of of XBP1S is higher than that of XBP1U.
