IL-24 联合顺铂通过 mTOR/P70S6K 信号通路抑制宫颈癌淋巴 转移的研究中文摘要目的:探讨 mTOR/P70S6K 信号通路是否参与宫颈癌 siha 细胞淋巴转移，并用 pDC316h重组质粒 IL-24 基因联合顺铂是否通过该途径来发挥抑制作用。方法:把我们的小裸鼠进行宫颈癌成功造模后，将它们随机分为 6 组，每组 7 只:第一 组为 PBS 对照组、第二组为 pDC316hIL-24 组、第三、四组为低剂量、高剂量顺铂组、 第五六祖分别为 pDC316hIL-24+低剂量顺铂组和 pDC316hIL-24+高剂量顺铂组。期间 注意观察裸鼠一般状况及移植瘤生长大小状况，并估计淋巴结转移情况，实验结束后 采用 CK 角蛋白检测获取的淋巴组织有否癌浸润，对于淋巴组织中的 IL-24mRNA 的 表达采用 PCR 法进行检测，最后将转移淋巴组织中 mTOR 和 P70S6K 蛋白用免疫荧 光法进行检测，定量分析采用 western blot 印迹法。结果:①IL-24 基因成功转染及表达；②CK 角蛋白检测获取的各组淋巴组织有癌浸润；③对淋巴转移数的结果及分析，发现干预后各组有差异，其中基因联合化疗药物组的 淋巴结转移数明显低于 PBS 对照组（p0.05），而基因联合低、高剂量顺铂组的淋巴 转移数无统计学差异（p0.05）；④免疫荧光法成功检测到各组转移淋巴组织中 mTOR/P70S6K 的表达，采用 Western 印迹法进行定量分析，相比对照组，各组呈下 降趋势，差异有统计学意义（p0.05），其中联合组表达显著下降（p0.05），差异有 统计学意义，且对 mTOR 与 P70S6K 进行相关分析后发现两者之间的表达具有正向相 关性(r=0.968，p 0.001)。结论:IL-24+低剂量顺铂治疗能达到 IL-24+高剂量顺铂的治疗效果；hIL-24 联合低剂量I顺铂组对于宫颈癌 siha 细胞淋巴转移的抑制作用显著明显；IL-24 联合顺铂在体内通过抑制 mTOR／P70S6K 信号通路的活性促进细胞凋亡，从而抑制人宫颈癌 siha 细胞 的淋巴转移。关键词:宫颈癌；淋巴结转移；白细胞介素；顺铂；mTORIIResearch on Inhibition to Human Uterine Lymph Node Metastasis through mTOR/P70S6K Signaling Pathway with IL-24 and CisplatinAbstractObjective:TOinvestigatemTOR/P70S6Ksignalpathwaysareinvolved in cervical cancer of siha cell oflymphatic metastasis, andpDC316Hil-24 plasmid gene combinedcisplatinin the way to play a role ofinhibition.Methods:Nude mice xenograft models were established with human cervical cancer Siha cells，and then animal models were randomly divided into 6 groups with7 mice in each group: PBS, IL-24, low-dose DDP, high-dose DDP, IL-24 combined low dose DDP andIL-24 combined high-dose DDP, After the intervention, throughobserve the conditions of weights and estimate the lymph node metastasis of our mouse , using CK keratin wastested to whether lymphoid tissue was transplanted tumor lymphatic metastasis, In addition, there was a detection that the expression of mRNA of IL-24 in lymphoid tissue by PCR technique , Finally, the expression of mTOR and P70S6K were tested by immunofluorescence technique as well as western blot imprinting method to q
