最后，采用间接ELISA的方法，以商品化的原料乳中固有脂肪酶脂蛋白脂肪酶的单抗为对照，研究表明本研究得到的抗荧光假单胞菌脂肪酶的抗血清能特异性地检测到荧光假单胞菌脂肪酶，这说明抗荧光假单胞菌脂肪酶的抗血清的特异性。本论文的研究结果为后续用单抗测定原料乳中的荧光假单胞菌脂肪酶奠定了一定的基础。关键词：荧光假单胞菌脂肪酶；抗原；细胞融合；杂交瘤细胞；ELISAAbstractAsweallknow,undertheinfluenceofmilkingconditions,therawmilkisveryvulnerabletomicrobesaftermilkingmachinesandotherequipmentprocessed.Inthesubsequentcoolingprocess,thepsychrophilethatcanwithstandlowtemperaturewillsurvive,andbecomethepredominantstrainssoon,forreproductionandmetabolisminmilk.Althoughpsychrotrophicbacteriacanbekilledduringthesterilizationprocess,buttheheatresistancelipaseitssecretioncanmaintainthermalstabilitybecauseofitsspecialstructure,andstillremainactiveafterpasteurizedevenUHTsterilization,therebydecomposingthefatinmilk,causingspoilageofmilkanddairyproducts.However,thetraditionaldetectionmethodofpsychrotropHicbacteriatakesalongtime,andcannotreflectthecaseofpsychrotropHicbacteriainrawmilkquicklyandaccurately,thuscannotassesswhethertherawmilkisqualifiedandsuitableforprocessing,andthencausinghugelossesinproduction.Therefore,developingafastmethodofPsychropHiledetectionisneeded.StudieshaveshownthatthereisasignificantexistingrelationshipbetweenthenumberofpsychrotropHicbacteriaandtheactivityofheat-resistantlipasetheysecreted.SothepsychrotropHicbacteriainrawmilkcanbepredictedbymeasuringlipase.Thisstudyaimedtoestablishahybridomacelllinesthatsecretedmonoclonalantibodyanti-Pseudomonasfluorescenslipase,andlaythematerialfoundationoftheELISAmethodforofrapiddetectionofPseudomonasfluorescenslipase.Inthisstudy,putPseudomonasfluorescens,whichisthepredominantpsychrotropHicstrainsinrawmilk,astheantigen.Firstofall,apreliminarypurificationbyultrafiltrationmethodtolipase,whichmakethepurityoflipasereach73.4%,meettherequirementconcentrationoftheantigen.Thentheinfluenceonlipaseactivitywasexaminedatdifferenttemperaturesreactiontime,differentpH,differentCa2+concentration,theresultsshowedthattheheat-resistantlipaseinthe50℃waterbathfor30min,63℃waterbathfor30min,72℃waterbathfor20min,90℃waterbathfo
