GLP-1 类似物对糖尿病肾病大鼠肾组织PEDF、TGF-β1表达的影响摘要目的：通过GLP-1 类似物对糖尿病肾病大鼠肾组织PEDF、TGF-β1 表达的影响研究，探讨GLP-1 类似物对糖尿病肾病影响的可能机制。方法：1、制备DN实验模型,分为糖尿病肾病组、低剂量治疗组、高剂量治疗组。2、治疗6周后观察各组血糖、血脂、24h尿蛋白定量，同时观察肾脏病理形态学改变。3、采用免疫组化和RT-PCR观察肾组织PEDF、TGF-β1蛋白及基因表达的变化。4、统计学处理：所有数据均以均数±标准差X±S表示, 采用SPSS17.0 统计分析软件进行单因素方差分析，组间比较采用LSD法，P0.05为差异有统计学意义。结果：1、经Exendin-4治疗6周后，与糖尿病肾病组相比，低剂量治疗组和高剂量治疗组的血糖、血脂、24h尿蛋白定量明显改善(P0.05)。与低剂量治疗组相比，高剂量治疗组改善更明显(P0.05)。2、通过免疫组化和RT-PCR观察肾组织PEDF、TGF-β1蛋白及基因表达的变化显示：与糖尿病肾病组相比，低剂量治疗组和高剂量治疗组大鼠肾组织的PEDF表达明显上升，TGF-β1表达明显下降（P0.05）。与低剂量治疗组相比，高剂量治疗组大鼠肾组织的表达变化更明显(P0.05)。结论：1、GLP-1类似物对DN大鼠具有一定的肾脏保护作用。2、GLP-1类似物对DN的肾脏保护具有剂量依赖性。关键词：GLP-1类似物；糖尿病肾病；PEDF；TGF-β1EffectofGLP-1analoguesontheExpressionofPEDF,TGF-β1intheKidney of theRatswithDiabeticNephropathyAbstractObjective: TostudytheEffectofGLP-1analoguesontheExpressionofPEDF,TGF-β1intheKidneyoftheRatswithDiabeticNephropath.Methods:1、Theratsmodelsofdiabeticnephropathywereestablishedbyinjectingwithstreptozotocinintraperitoneally.S-Dratswererandomlydividedinto3groups:thediabeticnephropathgroup，Low-dosetreatmentgroupandHigh-dosetreatmentgroup.2、Theratsineachgroupwererespectivelytreated6weeksaftermodelEstablishment.Bloodglucose(BS)，lipidand24hurinaryprotein(24h Upr) were determined.Massonstaining wasusedtoobservemorphoLogicalchangesofrenaltissue.3、ImmunohistochemistryandRT-PCRwereusedtodeterminetheproteinandmRNAexpressionofPEDF,TGF-β1inrenaltissue.4、Statisticalanalysis:AllstatisticalanalysiswasperformedwithWindowsSPSS17.0,datearepresentedX±S.
