健脾理气方药抑制HepG2人肝癌细胞增殖及诱导其凋亡形态学研究 　　[摘要] 目的：探讨中药复方健脾理气治疗肝癌的作用机制。方法：采用血清药理学方法，制备药物血清。应用MTT法测定细胞增殖抑制率，通过电镜技术检测药物血清作用后对细胞凋亡的影响。结果：药物血清作用24 h，中药小剂量组未见明显抑制作用，其余各组均表现出不同程度的抑制作用，中药大剂量组抑制率达8.81%；作用48 h，中药小剂量组出现抑制作用，各药物血清组随着作用时间的延长抑制更明显，中药大剂量组抑制率达39.27%，抑制效应呈时间依赖性。药物血清作用48 h后，电镜观察，可见肝癌细胞出现典型的细胞凋亡的形态学改变。结论：健脾理气具有明显抑制人肝癌细胞增殖、诱导肝癌细胞凋亡的作用，提示抑制细胞增殖和诱导细胞凋亡是健脾理气方药治疗肝癌的重要作用机制之一。 　　[关键词] 健脾理气；肝癌；凋亡；电镜 　　[中图分类号] R735.7 [文献标识码]B [文章编号]1673-7210（2008）06（b）-027-03 　　 　　Study on apoptosis effect induced and proliferarion of hepatoma by Jianpi Liqi herbs on HepG-2 cells and its mechanism 　　XING Xiao-jing，ZHANG Li-hong， CAI Yu-wen* 　　（Liaoning University of Traditional Chinese Medicine，Shenyang 110032，China） 　　[Abstract] Objective:To research the mechanism of traditional chinese medicine Jianpi Liqi herbs in treating primary hepatocellular carcinoma. Methods:To prepare serum by serum pharmacological methods. To determine the cell generation inhibition ratio with MTT methods and to detect the apoptotic effect by electron microscope technique.Results:within 24 h the effect of the drug serum of each group was differently obvious except in low concentration. The inhibition ratio in high concentration reached 8.81%.Within 48 h，the low concentration group showed inhibition，which was more obvious with the time-ratio and the high concentration group reached 39.27%-whose inhibition effects was time-dependent. Under the electron microscope，it was seen that liver cancer cells strain changed to apoptosis. Conclusion: Theserum of Jianpi Liqi herbs had effects on generation inhibition of human primary hepatocellular carcinoma cells and inducing apoptosis. All evidences indicated that generation inhibition and inducing apoptosis was one of the most important mechanisms of Jianpi Liqi herbs in treating hepatocellular carcinoma. 　　[Key words] Jianpi Liqi herbs;Primary hepatocellular carcinoma; Apoptosis; Electron microscope 　　 　　肝癌是一种原发于肝细胞或肝内胆管上皮细胞的常见消化道肿瘤，其发病率高，手术切除率低，愈后差，其发病率与死亡率均呈逐年上升趋势。肝癌具有发病
