万方数据 万方数据 Induced apoptotic effects and its mechanism of Lycopene in K562 cell Major: Pharmacy Postgraduate: SHI Qian-ying Supervisor: FANG Qin ,WANG Yong-lin [Abstract] Objective: Chemical treatment is the main way of treatment in chronic myelogenous leukemia (CML).But the drug resistance appears from the promotion of clinical application . The recent studies found that lycopene has antioxidant effect, especially its anti-cancer effect. This topic uses molecular biology techniques ,providing experimental basis for lycopene in the treatment of CML to investigate the induced apoptotic effects and its mechanism of lycopene on K562 cells. Methods: Different concentrations of lycopene (0，20，40，60 μmol·L-1) were treated with K562 cell after 24 hours ,48 hours,72 hours , 0 μmol· L-1 lycopene as the control group which named Ly0 group,20 μmol·L-1 lycopene named the Ly1 group，40 μmol·L-1 lycopene named the Ly2 group ，60 μmol·L-1 lycopene named the Ly3 group ; Ly0 group , Ly1 group, Ly2 group, Ly3 group cellular proliferation inhibition rate were observed by MTT method in K562 cell was after 24,48,72 hours ; Ly0 group , Ly1 group,Ly2 group, Ly3 group Cell apoptosis rate in K562 was inspected by Annexin V/PI double staining method after 48 hours; The mRNA expression of Bcl-2 and Caspase-3 gene in K562 cell are Ly0 group , Ly1 group, Ly2 group, Ly3 groups treated by lycopene was determined by using RT-PCR method after 48 hours；The protein expression of Bcl-2 and Caspase-3 gene in K562 cell what are Ly0 group, Ly1 group, Ly2 group, Ly3 groups treated by lycopene were determined by using Western Blot method after 48 hours. Results: As control group，MTT method showed that the Ly0 group, Ly1 group, Ly2 group, Ly3 groups cell proliferation inhibition rate in K562 treated by lycopene was positive associated with concentrations of lycopene ； Annexin V/PI method showed that the Ly0 group , Ly1 group, Ly2 group, Ly3 groups cell apoptosis ra
