摘要各组小鼠眼表及淋巴结中均有免疫炎症的改变，且炎症因子IFN—Y(Thl相关)、 摘要 各组小鼠眼表及淋巴结中均有免疫炎症的改变，且炎症因子IFN—Y(Thl相关)、 IL一6(Thl7相关)、IL一17(Thl7相关)表达升高，IL．13(T112相关)表达降低。 结论：研究发现在稳定的正常环境(温度23±1℃，相对湿度45±5％)条件下， 使用浓度为O．075％的BAC溶液每天滴眼两次(9am，9pm)，连续7天，可成功诱导 C57BL／6小鼠干眼模型。此模型能够很好模拟干眼病人的眼表体征，且免疫炎症改 变也与干眼病人相似。通过局部用药诱导不易产生全身副作用、诱导方法简单且诱 导周期短易推广，因此可以用于干眼病理机制的研究以及干眼治疗新药物的评估等。 关键词：干眼；苯扎氯铵；小鼠模型；炎症 II 万方数据 AbstractAbstract Abstract Abstract objective：To explore the optimal concentration of Benzalkonium Chloride used in the induction of dry eye model in C57BL／6 mice，and to detected the inflammatory changes in the ocular surface and cervical lymph nodes in this murine dry eye model Methods：The experimental C57BL／6 mice were randomly divided into six groups control group which received vehicle control eye drops．0．05％BAC mice which received 0．05％BAC eye drops，O．075％BAC mice which received O．075％BAC eye drops，O．1％ BAC mice which received O．1％BAC eye drops，O．1 5％BAC mice which received 0．1 5％ BAC eye drops and 0．2％BAC mice which received 0．1％BAC eye drops．All the mice were housed in stable environment(temperature of 23-4-1℃，relative humidity of 45士 5％)Topical application of BAK vehicle eye drops was performed twice day(9 am and 9 pm)for 7 days．The tear production in mice was by phenol red coaom test after 7 days．The corneal barrier function of mouse was assessed by OGD staining Immunofluorescence staining was used to evaluate apoptosis，proliferation，squamous metaplasia and MMPS in ocular surface．The number ofmice conjunctival goblet cells was tested by periodic acid-Schiff(periodicacid—Schiff,PAS)staining．In addition，real—time quantitative PCR(quantitative Real—time PCR)was used to test the expression levels of inflammatory cytokines such TNF—a，IFN吖，IL一1 7，IL-6，IL-1 3 and IL一1 13 in the conjunctiva and cervical lymph nodes Results：This study showed that，when the concentration of BAC higher than equal to 0．1％．corneal neovascularization and／or corneal ulcers appeared large proportion of C57BL／6 mice f2 eyes in 10 eye
