:Exosomesarereleasedmembranevesiclesderivedfromlateendosomes,whichsharestructuralandbio-
chemicalcharacteristicswithproteasomeosomesareresponsiblefortraffckingproteins,microRNAs(miRNAs),
andlongnon-codingRNAs(lncRNAs)amongcellsandregulatingvariouscellularprocesses,suchasdifferentiation,
proliferation,migration,invasion,andapoptosis.Althoughourknowledgeoftherolesofexosomesintheinitiation
andprogressionofleukemiaislimited,somestudieshaveindicatedthatexosomescanencompassmanyfunctional
factorswithanappropriatesortingsignal,therebysupportingthemetastasis,drugresistance,andimmuneescape
ofleukemiacells.Thisreviewinitiallyfocusesonthebiogenesisandcompositionofexosomesandthensumma-
rizestheapplicationofexosomesasascreeningbiomarkerandapotentialtherapeutictargetinleukemia.Many
recentreportsonthefunctionsofexosomesreleasedfromleukemiacellsarealsodiscussed,includingdrugresis-
tance,immunedysfunction,andmicroenvironmentmanipulation.Giventhecriticalrolesofexosomesinleukemia,
understandingthemechanismsregulatingthecompositionsandlevelsofexosomes,aswellasdefningexosome
functions,willultimatelyprovideadditionalinsightsintotheuseofexosomesastherapeuticagentsforleuk
