本實驗乃將微脂粒作為傳送載體的特性,以微脂粒包覆技術包覆蠶絲蛋白,利用,探討其平均粒徑大小、包覆率安定性,再進一步以進行皮膚保濕測試、皮膚彈力效果評估、皮膚過敏性及安全性之研究實驗結果不論是4℃或室溫環境下,都相較於薄膜/超音波震盪結果This experiment is to utilize the features of liposomes as a delivery vector and use liposomes coating technology to coat silk protein. We use different preparation methods and lipid composition to estimate the average particle size, encapsulation rate and stability, and further conduct the skin moisturizing test, skin elasticity effect evaluation test, and skin allergies and safety study.
Compared from the experiment results with different preparation methods, those liposomes prepared by alcohol injection, with a smaller average particle size, better encapsulation rate. But in terms of stability, either stored at 4 ℃ or room temperature, those are less stable when compared to the liposomes prepared by the film / sonication method.
These results indicate that different preparation methods and lipid compositions will affect the particle size of liposomes. For liposomes prepared by these two different methods, the liposomes prepared by ethanol injection method are with smaller particle size, but these two different preparation methods do have a common property, in both lipid composition, when cholesterol content increased, the greater the size of liposomes formed. Those lipid compositions which are added negatively charged lipids result in relatively smaller size, due to the increase of charged lipids inducing repulsive force between particles, and reducing the aggregation and fusion between particles, thus reducing the liposomes particle size.
