维普资讯 · 9O8 · ChinOEhthalRes，December2007 Vo1．25，No．12 ， · 实验研究 · TNF—OL对甲状腺相关眼病分化后眼眶脂肪细胞瘦素 mRNA表达的影响唐建罗清礼何为民 EffectofTNF-0con theexpression ofleptin mRNA in orbitaldifferentiated adipocytesofthyroidassociated ophthalmopathy TangJian，LuoQingli，HeWeimin．Departmentofophthalmology，WestChinaHospital，SichuanUniversity， Chengdu610041，China Abstract Objective Thyroidassociatedophthalmopathy(TAO)isanautoimmunitydiseasewithhypertrophyofocular muscleandincreaseofocularfat．Itisknown thatmanyfactorsareinvolvedin itspathogenesis，butinfluenceofleptin ingenesis anddevelopmentofTAO remainunclear．Thisstudywascu~iedouttoinvestigatetheeffectofTNF—dontheexpressionofleptin mRNAinorbitaldifferentiatedadipocytesofthyroidassociatedophthalmopathy(TAO)subjects． Methods Preadipocytesfrom orbitaltissuesofTAO patientsafterorbitaldecompressionwereculturedandcommittedtodifferentiate．Thedifferentiatedorbital adipocyteswereincubatedwith0．5，5，50ng／mL ofTNF—dfor24hoursor5ng／mLTNF—dofr0，12，24，48hours．Theexpression ofleptinmRNA in thedifferentiatedadipocyteswasdetected byreal—timefluorescentquantitativePCR． Results Induced preadipocytesdifferentiated intothecellswithmorphologicalfeatureofmatureadipocyteundertheappropriate in vitrocondition withtheredstainingofroilred．Aftertreatedwith 0．5—50mg／LTNF—d．theexpression ofleptinmRNA in thedifferentiated adipocyteswassignificantlysuppressedincomparisonwithcontrolgroup(F=10．401，P0．05)．TheexpressionofleptinmRNA inthe differentiated adipocytes was significantly decreased in 12．48 and 72 hoursafter5ng／mL ofTNF—d treatmentin comparisonwithbeforetreatment(F=28．834，P0．O1)． Conclusion TNF—dcansuppresstheexpressionofleptin
