Ab stra ct Aim:To optimize the preparation ofprocationic liposomes by using central composite design and investigate
the related characteristics of the procationic liposomes. Meth ods : Blank procationic liposomes were prepared by film
dispersion2filteration. The effects ofprotamine/ DNA ratio (w/w), CHETA molar percent (mol %) andCHETA/ DNA
ratio (w/w) on themean particle size, and thetransfection efficiencywere investigated. A second2order polynomial e2
quation was fittedto the data and theresultantmodelwas used to predict theresponse in the optimal region. Estimation
of the transfection efficiencywas performed by galactosidase assay inHepG2 cells using Lac as thereporter gene. Re2
sults: All the observed responsevariableswere found to be highly dependent on the formulation variables. Itwas found
