西格列汀联合吡格列酮对糖尿病大鼠肾脏TIMP-1、TGF-β1 表达的影响 熊盛池1，谢 燕，熊 利2 （1.四川省凉山州第一人民医院内分泌科，西昌 615000； 2. 四川省喜德县两河口中心卫生院内科，西昌 615000） 【摘要】目的:研究西格列汀联合吡格列酮对糖尿病大鼠肾脏的保护作用及其机制。方法：建立STZ（链脲佐菌素）诱导的糖尿病（Diabetes MellitusDM）SD大鼠模型，将大鼠随机分成4个组：组、组、西格列汀+吡格列酮组，设正常对照。组组、西格列汀+吡格列酮组组(P005)，以西格列汀+吡格列酮组(P0.01)。结论：西格列汀对DM大鼠的肾脏具有保护作用，而与吡格列酮联用效果更佳。其保护机制与TIMP-1和TGF-β1的在肾组织的表达减少有关。 【关键词】西格列汀；糖尿病肾病；基质金属蛋白酶组织抑制因子1；转化生长因子β1. 【中图分类号】R587.1 【文献标识码】A 【收稿日期】2012-06-29 Effects of Sitagliptin on the expression of TIMP-1 and TGF-β1 on kidney of diabetic Rats. Xiong Sheng-chi, et al （Department of endocrinology, First Hospital of Liang Shan，615000 China ） 【Abstract】Objective: To study the protective effects of the Sitagliptin on the kidney of diabetic rats.Methods:The model of DM rats induced by STZ were randomly divided into the following four groups: Sitagliptin group, diabetic model group, Sitagliptin combined with pioglitazone groupe , Pioglitazone group and normal control group. For each group of rats were respectively intervened for 12 weeks by the corresponding measures . and renal histopathological change was observation by Light microscopic. TIMP-1.TGF-β1 in renal tissue was detected by immunohistochemical technique. Results: The renal tissue histopathological of diabetic model group was markedly different from others group. The expression of TIMP-1 and TGF-β1 on renal cortex was increased in diabetic rats model group than others, Conclusion: Sitagliptin can protect the kidney from diabetic rats injury. And the protective effects of Sitagliptin combined with pioglitazone group is the most pronounced in all group .The protective mechanism in the kidney of Sitagliptin combined with pioglitazone may be associated with reduction the expression of TIMP-1 and TGF-β1 in renal tissue. 【Key words】 Sitagliptin; Diabetic ; Tissue inhibitor of matrix metalloproteinases-1;transforming growth factor β1. 西列汀为二肽基肽酶-4（dipeptidyl peptidase-4 DPP-4）抑制剂，DPP-4抑制剂是近年来上市的一类全新的突破性创新口服降糖药物，它能够提高一种被称为“肠促胰岛激素”:胰升血糖素样肽-1（GLP-1）和
