补虚化瘀法对产后抑郁大鼠雌激素和额叶p-CREB及p-ERK1/2的影响 谢萍*，冯俭*， （成都中医药大学附属医院，四川 成都610072） 摘要： 目的 探讨补虚化瘀方药参归仁合剂对产后抑郁（PPD）大鼠模型雌激素（E2）水平，额叶环磷腺苷反应元件结合蛋白(CREB), 细胞外调节蛋白激酶(ERK1/2)蛋白磷酸化水平表达的影响。方法 SD大鼠25只，随机分为正常对照组、模型组、参归仁高剂量组（14g·kg-1）、参归仁低剂量组（3.5g·kg-1）、左洛复（盐酸舍曲林）对照组（4.5mg·kg-1）。采用灌胃给药，连续30天。分别取血、额叶组织，采用放射免疫法检测大鼠E2水平，免疫印迹（Western blot,WB）检测额叶p-CREB、p-ERK1/2的表达水平。结果 与正常组相比，模型组大鼠血清E2，p-CREB，p-ERK1/2表达水平明显降低。与模型组相比，参归仁高、低剂量组，左洛复组额叶p-CREB表达量明显升高，参归仁低剂量组、左洛复组额叶p-ERK1/2表达量升高。结论 参归仁合剂能够明显提高PPD大鼠E2水平，促进额叶p-CREB，p-ERK1/2表达上调。 关键词：补虚化瘀法；雌激素；产后抑郁； CREB；ERK1/2；磷酸化水平 Effects of supplement deficiency and removal blood stasis on estrogen, p-CREB and p-ERK1/2 in postpartum depression model rats XIE Ping1*， HOU Yumin2，FENG Jian1* （1.The Hospital of Chengdu University of TCM，Chengdu，Sichuan，610072 P. China） Abstract： OBJECTIVE To investigate the effects of supplement deficiency and remove blood stasis on estrogen(E2), phosphor cAMP-response element binding protein (p-CREB )and phosphor extracellular?regulated?protein?kinases(p-ERK) in postpartum depression model rats. METHODS Twenty-five male Sprague Dawley rats were randomly divided into 5 groups: normal group, model group, high dose shenguiren treated group(14g·kg-1), low dose shenguiren treated group(3.5g·kg-1) and sertraline hydrochloride(4.5mg·kg-1)group. The rats accepted 30 days intragastric administration, and then drawn blood taken frontal lobe of brine. Analysis the level of serum estrogen by radioimmunoassay, phosphor CREB,ERK1/2 in frontal lobe of brine by western blot. RESULTS compared with the normal group, the level of the E2 p-CREB,p-ERK1/2 in the model group were decreased significantly. Compared with the model group, the expression of p-CREB in high, low dose shenguiren treated groups and sertraline hydrochloride group were increased significantly; and so did p-ERK1/2 in low dose shenguiren treated group, sertraline hydrochloride group.
