Scented rice is gaining wider popularity among consumers worldwide and its market price is twice higher than that of nonfragrant rice.Thus, it is very necessary to conduct a detailed research on the genetic basis underlying rice fragrance and metabolism of the major fragrant component, 2-acetyl-1-pyrroline 2AP . Genetic analysis shows that a single recessive gene fgr on chromosome 8 is associated with rice fragrance. In our previous work, the badh2 gene was finally identified as the fgr gene. Badh2 was found to be transcribed in all tissues tested except for roots, and the transcript was detected at higher abundance in young, healthy leaves than in other tissues. Based on the previous results, further analysis was carried out to elucidate the mechanism of rice fragrance gene and the component flavor 2AP formation. Multiple Badh2 transcripts were detected in RACE, RT-PCR and Northern blot hybridization, in which the complete full-length Badh2 transcript was much less abundant than partial Badh2 transcripts. However, 2AP levels were significantly reduced in cauliflower mosaic virus 35S-driven transgenic lines expressing the complete, but not the partial, Badh2 coding sequences. In accordance, the intact full-length BADH2 protein 503 residues appeared exclusively in nonfragrant transgenic lines and rice varieties, but not the fragrant ones. These results indicate that the full-length BADH2 protein encoded by Badh2 renders rice non-fragrant by inhibiting 2AP biosynthesis. Sequence alignment indicated that the 5846bp Badh2 gene containing 15 exons and 14 introns, whereas, the badh2 allele shows 8-bp deletion and three single nucleotide polymorphisms SNPs in its exon 7.
