类芽孢杆菌产絮凝多糖发酵条件优化及成分分析 姜彬慧, 李若男, 李凤达, 赵 研, 胡筱敏 东北大学资源与土木工程学院,辽宁 沈阳 110004 摘要:为提高微生物絮凝多糖的产量,应用响应面方法对一株高效产絮类芽孢杆菌(以下称A9)发酵产絮凝多糖的影响因素进行分析. 结果表明:①根据P-B(Plackett-Burman)法确定,影响A9产絮凝多糖的显著因子为装瓶量、ρ(MgSO4·7H2O)和ρ(可溶性淀粉);②通过Box-Behnken试验设计及响应面分析,确定了优化的培养基组成与培养条件,在ρ(可溶性淀粉)为17.18 g/L,ρ(酵母膏)为3 g/L,ρ(K2HPO4)为6 g/L,ρ(MgSO4·7H2O)为0.21 g/L,ρ(NaCl)为0.1 g/L,装瓶量为51.03 mL/250mL,pH为8的优化条件下,絮凝多糖实际产量为2.49 g/L,与理论预测值(2.50 g/L)接近;③利用红外光谱法检测A9产絮凝多糖的特征基团分别为—OH、—COO-、—C—O—C—和NHCOCH3等极性基团;④结合A9对不同碳源利用的单因素试验结果,推测A9产絮凝多糖的主要成分分别为含有α-吡喃型糖苷键的甘露糖和葡萄糖、酸性多糖和乙酰氨基多糖等. 关键词:絮凝多糖;响应面法;发酵条件优化;类芽孢杆菌 中图分类号:Q89 文献标识码A 文章编号:1001-6929(0000)00-00-00 Fermentation Conditions Optimization and omposition Analysis of Polysaccharide-based Bioflocculant Produced by Paenibacillus sp. Abstract: In order to increase the microbial flocculating polysaccharide production by Paenibacillus sp. A9, a novel microbial strain isolated from soils in Shenyang, the Plackett-Burman (P-B) design and Response Surface Methodology (RSM) were applied to optimize the influence factors in the fermentation culture process based on single factor experiments. The P-B results showed that the soluble starch, MgSO4·7H2O and amount of media were the three main factors that significantly affecting the polysaccharide production by Paenibacillus sp. A9 in the laboratory. A second order model of the polysaccharide production by strain A9 was obtained by Box-Behnken design and RSM analysis. Results showed that the optimal production of flocculating polysaccharide condition was 17.18g/L of soluble starch, 3g/L of yeast extract, 6g/L of K2HPO4,, 0.21g/L of MgSO4·7H2O, 0.1g/L of NaCl, 51.03mL/250mL of media amount and pH 8.0.
