石河子大学本科毕业论文 王芳 海岛棉GbMFT2基因在烟草中的遗传转化 PAGE \* MERGEFORMAT32 本科毕业论文 海岛棉GbMFT2基因在烟草中的 遗传转化 海岛棉GbMFT2基因在烟草中的遗传转化 学生：王芳 指导老师：黄先忠 摘要：已经证明，被子植物共享一个保守的磷脂酰乙醇胺结合（PEBP）结构域来调控开花时间。PEBP家族包括FT-like，TFL1-like和MFT-like三个亚家族，进化分析表明MOTHER OF FT AND TFL1 (MFT)是所有PEBP基因的祖先[10]。蛋白质序列比对发现GbMFT2蛋白的羧基端含有MFT蛋白都含有的脯氨酸，系统进化树分析表明GbMFT2编码产物与拟南芥AtMFT蛋白亲缘关系较近，他们属于同一分支。用不同浓度的ABA处理棉花种子后GbMFT2基因表达变化明显，表明GbMFT2基因的表达受ABA的调节。为了进一步研究该基因的功能，将过量表达载体p35S:GbMFT2叶盘法转化烟草，获得了7株T1代GbMFT2转基因烟草。种植T2代转基因烟草，发现转基因植株的开花时间均晚于非转基因植株。并用不同浓度NaCl胁迫处理T2代转GbMFT2基因烟草幼苗，发现转基因烟草的耐盐性明显高于野生型烟草。进一步分析GbMFT2基因的功能，以便可以利用该基因调节作物的开花时间及耐盐性。 关键词：PEBP家族；GbMFT2基因；叶盘法；晚花；耐盐 Genetic Transformation of Gossypium barbadense GbMFT2 in tobacco Abstract:Angiosperm sharing a conserved phosphatidylethanolamine-bindi- ng (PEBP) domain have been shown to be involved in the control of flowering time.The family is divided into three subfamilies, FT-like, TFL1-like and MFT-like. Evolution analysis showed that MOTHER OF FT AND TFL1 (MFT) is the ancestral form of all PEBP genes in plant, GbMFT2 gene has a PEBP conserved motif. In consistency with almost all MFT-like proteins, GbMFT2 has a conserved proline residue near the C-terminussimilarities similar with AtMFT and they all belong to the same clade. Exogenous abscisic acid (ABA) of different concentrations resulted in significant change in GbMFT2 expression in germinating seeds, indicating that the expression of this gene is regulated by ABA. To further study the function of the gene, I transformed the over-expression vector of p35S:GbMFT2 into tobacco. 7 p35S:GbMFT2 transformed tobacco plants were acquired. Analysis showed that the p35S:GbMFT2 transformed tobacco showed latter flower phenotype than non-transgenic wild type plants. Besides, through salt tolerance analysis I found that the p35S:GbMFT2 transformed tobacco showed stronger salt tolerance than non-transgenic wild type plants.
