The preparation is known as a fixed smear, which is then stained. ;Reagents: 1. Crystal violet 0.5% in distilled water. 2. Gram’s iodine 1 g iodine 2 g potassium iodide 300 ml distilled water ;Procedure: 1. On a clean slide place a loopful of water . 2. Transfer a very small amount of bacteria from one of the colonies you have selected using a sterile inoculating needle . 3. Spread bacteria over an area about the size of a dime to get a thin film. Sterilize needle immediately after making smear .;Procedure: 4. Allow slide to air dry. 5. Pass slide through flame, film side up, two or three times. 6. Cover slide with crystal violet, letting it remain 15 sec. Wash stain off with a gently flowing stream of tap water. 7. Cover with iodine solution for 30 sec. Wash with water.;Procedure: 8. Cover with acetone, agitate slide gently for 10 sec or until no more color is removed from smear. Wash with water. 9. Counterstain with safranin for 30 sec, and wash with water. 1
