[Methods] SD rats were obtained under sterile conditions, bone marrow, using density gradient centrifugation of rat MSCs, pass three generations of MSCs using double-labeled immunohistochemistry identified, were randomly divided into 4 groups: the concentration of 5, 10,15 μ mol / L of 5  Aza group and blank control group, using tetrazolium salt (MTT) assay different concentrations of 5  Aza on the growth and proliferation of MSCs; to contain 5,10,20 μ mol/L5  Aza medium induced cultured MSCs 24 h after switching to complete medium to continue to foster, using immunohistochemical staining were identified by 5  Aza induced differentiation of 3,7,14 d after the cell differentiation-related protein in muscle myosin (myosin) and desmin (desmin) staining. [Conclusion] The high concentration of 5  Aza inhibited the proliferation of MSCs in vitro; suitable concentration of 5  Aza can induce MSCs to differentiate into muscle cells, directed differentiation.
