Methods of antibiotics in agar diffusion within the use of tube plate method, culture temperature, the minimum detection limit of penicillin and the experimental strains ATCC941a streptomycin sulfate conditions, such as the maximum tolerated dose study. The results of incubation temperature is determined to be 28 ℃, the minimum detection limit of penicillin 0.008u/mg, experimental strains ATCC941a streptomycin sulfate maximum tolerated dose of 60mg/ml. Methods The parameters including culture temperature, limit of detection (LOD) of penicillin and the maximum tolerance dose of experimental strain ATCC9341a to streptomycin sulfate were detected by cylinder  plate method. Results The culture temperature was 28 ℃ and the LOD of penicillin was 0.008 u / ml, and the maximum tolerance dose of experimental strain ATCC9341a to streptomycin sulfate was 60 mg / ml. Conclusion This method was sensitive and can be used for penicillin residue assay.
