Psoralen on the A375 human melanoma cell proliferation and melanin synthesis Of: Tang Xiaoqin, Yan Ming, GAO Li, Peng Xiaoming, Hu Jiahui [Abstract] Objective To investigate serum levels of psoralen on the A375 with cell proliferation and function of melanin synthesis. Methods A375 human melanoma cells in vitro, in the logarithmic growth phase with different concentrations of psoralen in serum, MTT method and NaOH were used to detect cell proliferation and synthesis of black pigment. Results psoralen high, medium and low-dose group than in the blank serum containing serum significantly promoting proliferation in the concentration of 20%, high , medium and low doses of serum containing 3 group as the dose increases, melanin content was significantly higher (P lt;0.01). Conclusion psoralen containing serum can promote the proliferation of A375 melanoma cells, and to A375 human melanoma cells to increase melanin synthesis. Method A375 human melanoma cells were cultured in vitro. Different concentrations of drug-contained serum were added to the culture medium during logarithmic growth phase. Melanocyte culture was determined by MTT and NaOH method. Results There was a significant difference between the control and the drug-contained serum test group on proliferation, and there was significant increase in 20% serum drug-contained serum on melanogenesis (P lt;0.01). Conclusion Serum containing Psoralea has strong effect of promoting A375 human melanoma cells proliferation and melanogenesis in vitro.
