Anti-ObesityEffectofLactobacillusgasseriBNR17in High-SucroseDiet-InducedObeseMice Ji-HeeKang*,Sung-IlYun,Mi-HeePark,Jun-HongPark,So-YoungJeong,Han-OhPark RDcenter,BioneerCorporation,Daejeon,RepublicofKorea Abstract Previously, we reported that Lactobacillus gasseri BNR17 (BNR17), a probiotic strain isolated from human breast milk, inhibited increases in body weight and adipocyte tissue weight in high-sucrose diet-fed Sprague-Dawley (SD) rats and reducedglucoselevelsintype2diabetesmice.Inthecurrentstudy,weconductedfurtherexperimentstoextendthese observations and elucidate the mechanism involved. C57BL/6J mice received a normal diet, high-sucrose diet or high- sucrosedietcontainingL.gasseriBNR17(109or1010CFU)for10weeks.TheadministrationofL.gasseriBNR17significantly reducedthebodyweightandwhiteadiposetissueweightregardlessofthedoseadministered.InBNR17-fedgroups,mRNA levels of fatty acid oxidation-related genes (ACO, CPT1, PPARa, PPARd) were significantly higher and those of fatty acid synthesis-related genes(SREBP-1c,ACC)werelower comparedtothehigh-sucrose-diet group.TheexpressionofGLUT4, mainglucosetransporter-4,waselevatedinBNR17-fedgroups.L.gasseriBNR17alsoreducedthelevelsofleptinandinsulin inserum.Theseresultssuggestthattheanti-obesityactionsofL.gasseriBNR17canbeattributedtoelevatedexpressionof fattyacidoxidation-relatedgenesandreducedlevelsofleptin.Additionally,datasuggestedtheanti-diabetesactivityofL. gasseriBNR17maybetodueelevatedGLUT4andreducedinsulinlevels.
