CholinergicPlasticityofOscillatingNeuronalAssemblies inMouseHippocampalSlices MauraM.Zylla,XiaominZhang,SusanneReichinnek,AndreasDraguhn,MartinBoth* InstituteofPhysiologyandPathophysiology,UniversityHeidelberg,Heidelberg,Germany Abstract Themammalianhippocampusexpressesseveraltypesofnetworkoscillationswhichentrainneuronsintotransientlystable assemblies.Thesegroupsofco-activeneuronsarebelievedtosupporttheformation,consolidationandrecallofcontext- dependentmemories.Formationofnewassembliesoccursduringtheta-andgamma-oscillationsunderconditionsofhigh cholinergicactivity.Memoryconsolidationislinkedtosharpwave-rippleoscillations(SPW-R)duringdecreasedcholinergic tone.Wehypothesizedthatincreasedcholinergictonesupportsplasticchangesofassemblieswhilelowcholinergictone favors their stability. Coherent spatiotemporal network patterns were measured during SPW-R activity in mouse hippocampalslices.Wecomparedneuronalactivitywithintheoscillatingassembliesbeforeandafteratransientphaseof carbachol-induced gamma oscillations. Single units maintained their coupling to SPW-R throughout the experiment and couldbere-identifiedafterthetransientphaseofgammaoscillations.However,thefrequencyofSPW-R-relatedunitfiring wasenhancedaftermuscarinicstimulation.Atthenetworklevel,thesechangesresultedinalteredpatternsofextracellularly recordedSPW-Rwaveforms.Incontrast,recordingofongoingSPW-Ractivitywithoutintermittentcholinergicstimulation revealedremarkablystablerepetitiveactivationofassemblies.Theseresultsshowthatactivationofcholinergicreceptors inducesplasticityatthelevelofoscillatinghippocampalassemblies,inlinewiththedifferentroleofgamma-andSPW-R networkactivityformemoryformationand–consolidation,respectively.
