Methodology/PrincipalFindings:WeprofiledsixhistonemodificationsalongsideRNApolymeraseII(PolII)andCTCFintwo cell lines, HeLa S3 and NTERA-2 clone D1 (NT2/D1), by chromatin immunoprecipitation using an in-house spotted DNA array,constructedwith1.8kboverlappingplasmidclones.Inbothcells,morethan90%oftranscriptionstartsites(TSSs)of expressedgenesshowedenrichmentswithPolII,di-methylatedlysine4ofhistoneH3(H3K4me2),tri-methylatedlysine4of histoneH3(H3K4me3)oracetylatedH3(H3Ac),whereasmono-methylatedlysine4ofhistoneH3(H3K4me1)signalsdidnot correlatewithexpression.NoTSSswereenrichedwithtri-methylatedlysine27ofhistoneH3(H3K27me3)inHeLaS3,while eightTSSs(4expressed)showedenrichmentsinNT2/D1.WehavealsolocatedseveralCTCFbindingsitesthatarepotential insulatorelements. Conclusions/Significance:Insummary,weannotatedanumberofputativeregulatoryelementsin20q13.12andwenton toverifyexperimentallyasubsetofthemusingdualluciferasereporterassays.Correlatingthisdatatosequencevariation canaididentificationofdiseasecausingvariants.
