Recentpapersshowedthatduringangiogenesis,AQP1isexpressedpreferentiallybymicrovessels,favoringangiogenesis viatheincreaseofpermeabilityInparticular,inAQP1nullmice,endothelialcellmigrationisimpairedwithoutalteringtheir proliferationoradhesion.Therefore,AQP1hasbeenproposedasanovelpromoteroftumorangiogenesis. Methods/Findings:UsingtargetedsilencingofAQP1geneexpression,animpairmentintheorganizationofF-actinanda reducedmigrationcapacitywasdemonstratedinhumanendothelialandmelanomacelllines.Interestingly,weshowed,for thefirsttime,thatAQP1co-immunoprecipitatedwithLin-7.Lin7-GFPexperimentsconfirmedco-immunoprecipitation.In addition, the knock down of AQP1 decreased the level of expression of Lin-7 and b-catenin and the inhibition of proteasomecontrastedpartiallysuchadecrease. Conclusions/Significance:Alltogether,ourfindingsshowthatAQP1playsaroleinsidethecellsthroughLin-7/b-catenin interaction.SucharoleofAQP1isthesameinhumanmelanomaandendothelialcells,suggestingthatAQP1playsaglobal physiologicalrole.Amodelispresented.
