Abstract
Itwasshownthatatlowconcentrationsguanidinehydrochloride(GdnHCl)cancauseaggregationofproteinsinpartially
foldedstateandthatfluorescentdye1-anilinonaphthalene-8-sulfonicacid(ANS)bindswiththeseaggregatesratherthan
withhydrophobicclustersonthesurfaceofproteininmoltenglobulestate.ThatiswhytheincreaseinANSfluorescence
intensity is often recorded in thepathway of protein denaturation by GdnHCl, but not by urea. So what was previously
believedtobethemoltenglobulestateinthepathwayofproteindenaturationbyGdnHCl,inreality,forsomeproteins
representstheaggregatesofpartiallyfoldedmolecules.
