Methodology/PrincipalFindings:Amoexhibitsauniqueexpressionpatternduringspermatogenesis.Inspermatocytes,Amois restrictedtotheendoplasmicreticulum(ER)whereasinmaturesperm,Amoclustersatthedistaltipofthespermtail.Herewe showthatflagellarlocalizationofAmoisrequiredforspermstorage.ThisraisedthequestionofhowAmoattherearendof spermregulatesforwardmovementintothestorageorgans.Inordertoaddressthisquestion,weusedinvivoimagingofdual labelledspermtodemonstratethatDrosophilaspermnavigatebackwardsinthefemalereproductivetract.Inaddition,weshow thatspermexhibithyperactivationupontransfertotheuterus.Amomutantspermremaincapableofreversemotilitybutfailto displayhyperactivationanddirectedmovement,suggestingthatthesefunctionsarerequiredforspermstorageinflies. Conclusions/Significance:Amoispartofasignallingcomplexattheleadingedgeofthespermtailthatmodulatesflagellar beatingandthatguidesabackwardspathintothestorageorgans.Ourdatasupportane
