Consistent with our proposition, the HAgel differs from agar by suppressing the typicalside-by-sidealignmentstoararepopulation.Examinationofbacterialsurfacemoleculesthatmaycontributetothe daughtercells’arrangementyieldedanobservationthat,withdisruptedlpp,theE.colidaughtercellsincreasinglyformed non-typicalpatterns,i.e.neitherslidingside-by-sideinparallelnorformingelongatedstrings.Therefore,ourresultssuggest stronglythattheearlycellpatterningisaffectedbymultipleinteractionfactors.Withoscillatoryopticaltweezers,wefurther demonstratedthattheinteractionforcedecreasedinbacteriawithoutLpp,aresultsubstantiatingournotionthattheside- by-sideslidingphenomenondirectlyreflectsthestrengthofin-situinteractionbetweenbacteriaandsubstratum.
