Resistance to pyrethroid insecticides in the malaria vector Anopheles gambiae is receiving increasing attention becauseitthreatensthesustainabilityofmalariavectorcontrolprogramsinsub-SaharanAfrica.Anunderstandingofthe molecularmechanismsconferringpyrethroidresistancegivesinsightintotheprocessesofevolutionofadaptivetraitsand facilitatesthedevelopmentofsimplemonitoringtoolsandnovelstrategiestorestoretheefficacyofinsecticides.Forthis purpose,itisessentialtounderstandwhichmechanismsareimportantinwildmosquitoes.Here,ouraimwastoidentify enzymesthatmaybeimportantinmetabolicresistancetopyrethroidsbymeasuringgeneexpressionforover250genes potentiallyinvolvedinmetabolicresistanceinphenotypedindividualsfromahighlyresistant,wildA.gambiaepopulation fromGhana.AcytochromeP450,CYP6P3,wassignificantlyoverexpressedinthesurvivors,andweshowthatthetranslated enzymemetabolisesbothalpha-cyanoandnon–alpha-cyanopyrethroids.Thisisthefirststudytodemonstratethecapacity ofaP450identifiedinwildA.gambiaetometaboliseinsecticides.Thefindingsaddtotheunderstandingofthegenetic basisofinsecticideresistanceinwildmosquitopopulations.
