Gas6DownregulationImpairedCytoplasmicMaturation andPronuclearFormationIndependenttotheMPF Activity Kyeoung-HwaKim,Eun-YoungKim,YunaKim,EunjuKim,Hyun-SeoLee,Sook-YoungYoon,Kyung-Ah Lee* DepartmentofBiomedicalScience,CollegeofLifeScience,FertilityCenter,CHAResearchInstitute,CHAUniversity,CHAGeneralHospital,Seoul,Korea Abstract Previously,wefoundthatthegrowtharrest-specificgene6(Gas6)ismorehighlyexpressedingerminalvesicle(GV)oocytes thaninmetaphaseII(MII)oocytesusingannealingcontrolprimer(ACP)-PCRtechnology.Thecurrentstudywasundertaken toinvestigatetheroleofGas6inoocytematurationandfertilizationusingRNAinterference(RNAi).Interestingly,despitethe specificandmarkeddecreaseinGas6mRNAandproteinexpressioninGVsafterGas6RNAi,nuclearmaturationincluding spindlestructuresandchromosomesegregationwasnotaffected.TheonlydiscernibleeffectinducedbyGas6RNAiwasa changeinmaturationpromotingfactor(MPF)activity.Afterparthenogeneticactivation,Gas6RNAi-treatedoocytesatthe 2+ MIIstagehadnotdevelopedfurtherandarrestedatMII(90.0%).AfterstimulationwithSr ,Gas6-silencedMIIoocyteshad markedly reduced Ca oscillation and exhibited no exocytosis of cortical granules. In these oocytes, sperm penetration 2+ occurredduringfertilizationbutnotpronucleus(PN)formation.Byroscovitineandcolcemidtreatment,wefoundthatthe Gas6knockdownaffectedcytoplasmicmaturationdirectly,independenttothechangedMPFactivity.Theseresultsstrongly suggestthat1)theGas6signalingitselfisimportanttothecytoplasmicmaturation,butnotnuclearmaturation,and2)the decreasedGas6expressionanddecreasedMPFactivityseparatelyormutuallyinfluencespermheaddecondensationand PNformation.
