GeneratingaKnockdownTransgeneagainstDrosophila HeterochromaticTim17bGeneEncodingMitochondrial TranslocaseSubunit MikaelGarabedian.,MichaelJarnik.,ElenaKotova,AlexeiV.Tulin* FoxChaseCancerCenter,Philadelphia,Pennsylvania,UnitedStatesofAmerica Abstract Heterochromaticregionsofeukaryoticgenomescontainmultiplefunctionalelementsinvolvedinchromosomaldynamics, aswellasmultiplehousekeepinggenes.Cytologicalandmolecularpeculiaritiesofheterochromaticlocicomplicategenetic studiesbasedonstandardapproachesdevelopedusingeuchromaticgenes.Here,wereportthedevelopmentofanRNAi- basedknockdowntransgenicconstructandredfluorescentreportertransgeneforasmallgene,Tim17b,whichlocalizesin constitutive heterochromatin of Drosophila melanogaster third chromosome and encodes a mitochondrial translocase subunit.WedemonstratethatTim17bproteinisrequiredstrictlyforproteindeliverytomitochondrialmatrix.Knockdownof Tim17b completely disrupts functions of the mitochondrial translocase complex. Using fluorescent recovery after photobleachingassay,weshowthatTim17bproteinhasaverystablelocalizationinthemembranesofthemitochondrial networkandthatitsexchangerateisclosetozerowhencomparedwithsolubleproteinsofmitochondrialmatrix.These resultsconfirmthatwehavedevelopedcomprehensivetoolstostudyfunctionsofheterochromaticTim17bgene.
