Methodology/PhysicalFindings:Inthisreportwehaveusedasimilarmethodtodirectinducedpluripotentstemcells(iPS) fromhumanfibroblaststoaretinalprogenitorfate,competenttogeneratephotoreceptors.Wealsofoundwecouldpurify thephotoreceptorsderivedfromtheiPScellsusingfluorescenceactivatedcellsorting(FACS)afterlabelingphotoreceptors withalentivirusdrivingGFPfromtheIRBPcis-regulatorysequences.Moreover,wefoundthatwhenwetransplantedthe FACS purified iPSC derived photoreceptors, they were able to integrate into a normal mouse retina and express photoreceptormarkers. Conclusions:ThisreportprovidesevidencethatenrichedpopulationsofhumanphotoreceptorscanbederivedfromiPS cells.
