We have previously shown that geranylgeranyl transferaseinhibition(donewithGGTI-298)stimulatesanti-melanomaimmuneresponsethroughMHC-Iandcostimulatory moleculeexpressionintheB16F10murinemodel[1]. Methodology/PrincipalFindings:Inthisstudy,itisshownthatvaccinationwithmIFN-gandGGTI-298pretreatedB16F10 cellsinducesaprotectionagainstuntreatedtumorgrowthandpulmonarymetastasesimplantation.Furthermore,usinga humanmelanomamodel(LB1319-MEL),wedemonstratedthatinvitrotreatmentwithhIFN-candGGTI-298ledtotheup regulationofMHC-IandacostimulatorymoleculeCD86anddownregulationofaninhibitorymoleculePD-1L.Co-culture experimentswithperipheralbloodmononuclearcells(PBMC)revealedthatmodificationsinducedbyhIFN-candGGTI-298 ontheselectedmelanomacells,enablesthestimulationoflymphocytesfromHLAcompatiblehealthydonors.Indeed,as comparedwithuntreatedmelanomacells,pretreatmentwithhIFN-candGGTI-298togetherrenderedthemelanomacells moreefficientatinducingthe:i)activationofCD8Tlymphocytes(CD8+/CD69+);ii)proliferationoftumor-specificCD8T cells(MelanA-MART1/TCR+);iii)secretionofhIFN-c;andiv)anti-melanomaspecificcytotoxiccells. Conclusions/Significance:ThesedataindicatethatpharmacologicaltreatmentofmelanomacelllineswithIFN-candGGTI- 298stimulatestheirimmunogenicityandcouldbeanovelapproachtoproducetumorcellssuitablefo
