MetagenomicComparisonofTwoThiomicrospira LineagesInhabitingContrastingDeep-SeaHydrothermal Environments WilliamJ.Brazelton*,JohnA.Baross SchoolofOceanographyandCenterforAstrobiologyandEarlyEvolution,UniversityofWashington,Seattle,Washington,UnitedStatesofAmerica Abstract Background: The most widespread bacteria in oxic zones of carbonate chimneys at the serpentinite-hosted Lost City hydrothermalfield,Mid-AtlanticRidge,belongtotheThiomicrospiragroupofsulfur-oxidizingchemolithoautotrophs.Itis unclear why Thiomicrospira-like organisms thrive in these chimneys considering that Lost City hydrothermal fluids are notablylackinginhydrogensulfideandcarbondioxide. Methodology/PrincipalFindings:HerewedescribemetagenomicsequencesobtainedfromaLostCitycarbonatechimney thatarehighlysimilartothegenomeofThiomicrospiracrunogenaXCL-2,anisolatefromabasalt-hostedhydrothermalvent inthePacificOcean.EventhoughT.crunogenaandLostCityThiomicrospirainhabitdifferenttypesofhydrothermalsystems indifferentoceans,theirgenomiccontentsarehighlysimilar.Forexample,sequencesencodingthesulfuroxidationand carbonfixationpathways(includingacarbonconcentrationmechanism)ofT.crunogenaarealsopresentintheLostCity metagenome.Comparativegenomicanalysesalsorevealedsubstantialgenomicchangesthatmusthaveoccurredsincethe divergenceofthetwolineages,includinglargegenomic rearrangements,genefusion events,a prophageinsertion, and transposaseactivity. Conclusions/Significance: Our results show significant genomic similarity between Thiomicrospira organisms inhabiting differentkindsofhydrothermalsystemsindifferentoceans, suggestingthattheseorganismsarewidespreadandhighly adaptable. These dataalso indicategenomic processes potentially associated withthe adaptationof these lineagesinto strikinglydifferenthabitats.
