Methodology/PrincipalFindings:UsingRACEassay,wecloneda1106bptranscriptnamedMyctarget1transcriptvariant 1(MYCT1-TV)andconfirmeditstranscriptionalstartsitewaslocatedat140bpupstreamoftheATGstartcodonofMYCT1- TV.Luciferase,electrophoreticmobilityshiftandchromatinimmunoprecipitationassaysconfirmedc-Myccouldregulatethe promoteractivityofMYCT1-TVbyspecificallybindingtotheE-boxelementswithin2886to2655bpregion.Theseresults were further verified by site-directed mutagenesis and RNA interference (RNAi) assays. MYCT1-TV and MYCT1 expressed lowerinLSCCthanthoseinpairedadjacentnormallaryngealtissues,andoverexpressionofMYCT1-TVandMYCT1could inhibitcellproliferationandinvasionandpromoteapoptosisinLSCCcells. Conclusions/Significance: Our data indicate that MYCT1-TV, a novel MYCT1 transcript, is regulated by c-Myc and down- regulationofMYCT1-TV/MYCT1couldcontributetoLSCCdevelopmentandfunction.
