Abstract
Background: Cyanosis and myocardial hypertrophy frequently occur in combination. Hypoxia or cyanosis can be potent
inducersofangiogenesis,regulatingtheexpressionofhypoxia-inducible factors(HIF),vascular endothelialgrowthfactors
(VEGF),andVEGFreceptors(VEGFR-1and2);incontrast,pressureoverloadhypertrophyisoftenassociatedwithimpairedpro-
angiogenic signaling and decreased myocardial capillary density. We hypothesized that the physiological pro-angiogenic
responsetocyanosisinthehypertrophiedmyocardiumisbluntedthroughdifferentialHIFandVEGF-associatedsignaling.
Methods and Results: Newborn rabbits underwent aortic banding and, together with sham-operated littermates, were
transferredintoahypoxicchamber(FiO2=0.12)at3weeksofage.Controlbandedorsham-operatedrabbitswerehousedin
normoxia.Systemiccyanosiswasconfirmed(hematocrit,arterialoxygensaturation,andserumerythropoietin).Myocardial
tissuewasassayedforlowoxygenconcentrationsusingapimonidazoleadduct.At4weeksofage,HIF-1aandHIF-2aprotein
levels,HIF-1aDNA-bindingactivity,andexpressionofVEGFR-1,VEGFR-2,andVEGFweredeterminedinhypoxicandnormoxic
rabbits.At6weeksofage,left-ventricularcapillarydensitywasassessedbyimmunohistochemistry.Undernormoxia,capillary
densitywasdecreasedinthebandedrabbitscomparedtonon-bandedlittermates.Asexpected,non-hypertrophiedhearts
responded to hypoxia with increased capillary density; however, banded hypoxic rabbits demonstrated no increase in
