Over-ExpressionofLSD1PromotesProliferation, MigrationandInvasioninNon-SmallCellLungCancer TangfengLv1,DongmeiYuan1,XiaohuiMiao1,YanlingLv1,PingZhan2,XiaokunShen1,YongSong1* 1DepartmentofRespiratoryMedicine,JinlingHospital,NanjingUniversitySchoolofMedicine,Nanjing,China,2FirstDepartmentofRespiratoryMedicine,NanjingChest Hospital,Nanjing,China Abstract Background:Lysinespecificdemethylase1(LSD1)hasbeenidentifiedandbiochemicallycharacterizedinepigenetics,but thepathologicalrolesofitsdysfunctioninlungcancerremaintobeelucidated.Theaimofthisstudywastoevaluatethe prognosticsignificanceofLSD1expressioninpatientswithnon-smallcelllungcancer(NSCLC)andtodefineitsexactrolein lungcancerproliferation,migrationandinvasion. Methods: The protein levels of LSD1 in surgically resected samples from NSCLC patients were detected by immunohistochemistry or Western blotting. The mRNA levels of LSD1 were detected by qRT-PCR. The correlation of LSD1expressionwithclinicalcharacteristicsandprognosiswasdeterminedbystatisticalanalysis.Cellproliferationratewas assessedbyMTSassayandimmunofluorescence.Cellmigrationandinvasionweredetectedbyscratchtest,matrigelassay andtranswellinvasionassay. Results:LSD1expressionwashigherinlungcancertissuemorethaninnormallungtissue.Ourresultsshowedthatover- expressionofLSD1proteinwereassociatedwithshorteroverallsurvivalofNSCLCpatients.LSD1waslocalizedmainlytothe cancercellnucleus.InterruptionofLSD1usingsiRNAorachemicalinhibitor,pargyline,suppressedproliferation,migration andinvasionofA549,H460and293Tcells.Meanwhile,over-expressionofLSD1enhancedcellgrowth.Finally,LSD1was showntoregulateepithelial-to-mesenchymaltransitioninlungcancercells. Conclusions:Over-expressionofLSD1wasassociatedwithpoorprognosisinNSCLC,andpromotedtumorcellproliferation, migrationandinvasion.TheseresultssuggestthatLSD1isatumor-promotingfactorwithpromisingtherapeuticpotential forNSCLC.
