Until now, only a few studies on sjTREC detection in bloodstain samples were reported, which were based on a small sample of subjects of a limited age range, although bloodstains are much more frequently encountered in forensic practice. In this present study, we adopted the sensitive Taqmanreal-timequantitativepolymerasechainreaction(qPCR)methodtoperformsjTRECquantificationinbloodstains fromindividualsrangingfrom0–86yearsold(n=264).TheresultsrevealedthatsjTRECcontentsinhumanbloodstainswere declinedinanage-dependentmanner(r=20.8712).TheformulaofageestimationwasAge =27.1815Y242.45869.42(Y dCtTBP-sjTREC;9.42standarderror).Furthermore,wetestedfortheinfluenceofshort-orlong-storagetimebyanalyzingfresh andstoredbloodstainsfromthesameindividuals.Remarkably,nostatisticallysignificantdifferenceinsjTRECcontentswas foundbetweenthefreshandoldDNAsamplesovera4-weekofstoragetime.However,significantloss(0.16–1.93dCt)in sjTRECcontentswasdetectedafter1.5yearsofstoragein31samples.Moreover,preliminarysjTRECquantificationfromup to20-year-oldbloodstainsshowedthatthoughthesjTRECcontentsweredetectableinallsamplesandhighlycorrelated withdonorage,atime-dependentdecreaseinthecorrelationcoefficientrwasf
