PresagingCriticalResiduesinProteininterfaces-Web Server(PCRPi-W):AWebServertoChartHotSpotsin ProteinInterfaces JoanSeguraMora.,SalamA.Assi.¤,NarcisFernandez-Fuentes* SectionofExperimentalTherapeutics,LeedsInstituteofMolecularMedicine,UniversityofLeeds,Leeds,UnitedKingdom Abstract Background:Itiswellestablishedthatonlyaportionofresiduesthatmediateprotein-proteininteractions(PPIs),theso- calledhotspot,contributesthemosttothetotalbindingenergy,andthusitsidentificationisanimportantandrelevant questionthathasclearapplicationsindrugdiscoveryandproteindesign.Theexperimentalidentificationofhotspotsis howeveralengthyandcostlyprocess,andthusthereisaninterestincomputationaltoolsthatcancomplementandguide experimentalefforts. PrincipalFindings:Here,wepresentPresagingCriticalResiduesinProteininterfaces-Webserver(http://www.bioinsilico. org/PCRPi),awebserverthatimplementsarecentlydescribedandhighlyaccuratecomputationaltooldesignedtopredict critical residues inproteininterfaces: PCRPi.PRCPidepends ontheintegration ofstructural, energetic,andevolutionary- basedmeasuresbyusingBayesianNetworks(BNs). Conclusions: PCRPi-W has been designed to provide an easy and convenient access to the broad scientific community. Predictionsarereadilyavailablefordownloadorpresentedinawebpagethatincludesamongotherinformationlinksto relevantfiles,sequenceinformation,andaJmolapplettovisualizeandanalyzethepredictionsinthecontextoftheprotein structure.
