PrevalenceandGeneticCharacterizationofPertactin- DeficientBordetellapertussisinJapan NaoOtsuka,Hyun-JaHan¤a,HiromiToyoizumi-Ajisaka,YukitsuguNakamura,YoshichikaArakawa¤b, KeigoShibayama,KazunariKamachi* DepartmentofBacteriologyII,NationalInstituteofInfectiousDiseases,Tokyo,Japan Abstract Theadhesinpertactin(Prn)isoneofthemajorvirulencefactorsofBordetellapertussis,theetiologicalagentofwhooping cough.However,asignificantprevalenceofPrn-deficient(Prn2)B.pertussiswasobservedinJapan.ThePrn2isolatewas firstdiscoveredin1997,and33(27%)Prn2isolateswereidentifiedamong121B.pertussisisolatescollectedfrom1990to 2009.SequenceanalysisrevealedthatallthePrn2isolatesharborexclusivelythevaccine-typeprn1alleleandthatlossof Prnexpressioniscausedby2differentmutations:an84-bpdeletionoftheprnsignalsequence(prn1DSS,n=24)andan IS481 insertion in prn1 (prn1::IS481, n=9). The frequency of Prn2 isolates, notably those harboring prn1DSS, significantly increasedsincetheearly2000s,andPrn2isolatesweresubsequentlyfoundnationwide.Multilocusvariable-numbertandem repeatanalysis(MLVA)revealedthat24(73%)of33Prn2isolatesbelongtoMLVA-186,and6and3Prn2isolatesbelongto MLVA-194andMLVA-226,respectively.The3MLVAtypesarephylogeneticallycloselyrelated,suggestingthatthe2Prn2 clinicalstrains(harboringprn1DSSandprn1::IS481)haveclonallyexpandedinJapan.Growthcompetitionassaysinvitroalso + demonstratedthatPrn2isolateshaveahighergrowthpotentialthanthePrn back-mutantsfromwhichtheywerederived. Ourobservationssuggestedthathumanhostfactors(geneticfactorsandimmunestatus)thatselectforPrn2strainshave arisenandthatPrnexpressionisnotessentialforfitnessundertheseconditions.
