Sallametal.AMBExpress2011,1:38 /content/1/1/38 ORIGINAL OpenAccess Productionoptimizationofcyanophycinase ChpEalfromPseudomonasalcaligenesDIP1 AhmedSallam,DimitarKalkandzhievandAlexanderSteinbüchel* Abstract PseudomonasalcaligenesDIP1producesanextracellularcyanophycinase(CphEal).Thecorrespondinggene(cphEal) wasidentifiedfromsubclonesofagenomicDNAgenelibrarybyheterologouslyexpressingthefunctionallyactive enzymeinEscherichiacoli.Thenucleotidesequenceofthegene(1260basepairs)wasdeterminedindicatinga theoreticalmassof43.6kDa(matureCphEal)plusaleaderpeptideof2,6kDawhichcorrespondswelltothe apparentmolecularmassof45kDaasrevealedbySDS.Theenzymeexhibitedahighsequenceidentityof 91%withtheextracellularcyanophycinasefromP.anguillisepticastrainBIandcarriedanN-terminalSecsecretion signalpeptide.AnalysisoftheaminoacidsequenceofcphErevealedaputativecatalytictriadconsistingofthe serinemotifGXSXGplusahistidineandaglutamateresidue,suggestingacatalyticmechanismsimilartoserine- typeproteases.Thecyanophycinase(CphEal)washeterologouslyproducedintwodifferentE.colistrains(Top10 andBL21(DE3))fromtwoplasmidvectors(pBBR1MCS-4andpET-23a(+)).ThesignalpeptideofCphEal wascleaved inE.coli,suggestingactiveexportoftheproteinatleasttotheperiplasm.Substantialenzymeactivitywasalso presentintheculturesupernatants.TheextracellularcyanophycinaseactivitiesinE.coliwerehigherthanactivities inthewildtypeP.alcaligenesDIP1incomplexLBmedium.Highestextracellularenzymeproductionwasachieved withE.coliBL21(DE3)expressingCphEal frompBBR1MCS-4.UsingM9minimalmediumwaslesseffective,butthe relativelylowcostofmineralsaltmediamakestheseresultsimportantfortheindustrial-scaleproductionof dipeptidesfromcyanophycin.
