RickAExpressionIsNotSufficienttoPromoteActin- BasedMotilityofRickettsiaraoultii PremanandBalraj,KhalidElKarkouri,GuyVestris,LeonEspinosa,DidierRaoult,PatriciaRenesto* Unite′ desRickettsies,URMITEIRD-CNRSUMR6236,Faculte′ deMe′decine,Marseille,France Abstract Background:RickettsiaraoultiiisanovelRickettsiaspeciesrecentlyisolatedfromDermacentorticksandclassifiedwithinthe spottedfevergroup(SFG).TheinabilityofR.raoultiitospreadwithinL929cellssuggeststhatthisbacteriumisunableto polymerizehostcellactin,apropertyexhibitedbyallSFGrickettsiaeexceptR.peacocki.Thisresultledustoinvestigateif RickA,theproteinthoughttogenerateactinnucleation,wasexpressedwithinthisrickettsiaspecies. Methodology/Principal Findings: Amplification and sequencing of R. raoultii rickA showed that this gene encoded a putative565aminoacidproteinhighlyhomologoustothosefoundinotherrickettsiae.Usingimmunofluorescenceassays, wedeterminedthatthemotilitypattern(i.e.microcoloniesorcell-to-cellspreading)ofR.raoultiiwasdifferentdependingon thehostcelllineinwhichthebacteriareplicated.Incontrast,underthesameexperimentalconditions,R.conoriisharesthe samephenotypebothinL929andinVerocells.Transmissionelectronmicroscopyanalysisofinfectedcellsshowedthat non-motilebacteriawerefreeinthecytosolinsteadofenclosedinavacuole.Moreover,western-blotanalysisdemonstrated thatthedefectofR.raoultiiactin-basedmotilitywithinL929cellswasnotrelatedtolowerexpressionofRickA. Conclusion/Significance: These results, together with previously published data about R. typhi, strongly suggest that anotherfactor,apartfromRickA,maybeinvolvedwithberesponsibleforactin-basedmotilityinbacteriafromtheRickettsia genus.
