The full genome of HPV-125 is 7,809-bp in length with a GC content of 46.4%. By comparing the nucleotide sequence of the complete L1 gene, HPV-125 is phylogenetically placed within cutaneotrophicspecies2ofAlphapapillomaviruses,andismostcloselyrelatedtoHPV-3andHPV-28.HPV-125hasatypical genomicorganizationofAlphapapillomavirusesandcontainsgenescodingforfiveearlyproteins,E6,E7,E1,E2andE4and twolatecapsidproteins,L1andL2.Thegenomecontainstwonon-codingregions:thefirstlocatedbetweentheL1andE6 genes(nucleotidepositions7,137–7,809,length673-bp)andthesecondbetweengenesE2andL2(nucleotidepositions 3,757–4,216,length460-bp).TheE6proteinofHPV-125containstworegularzinc-bindingdomainsataminoacidpositions 29and102,whereastheE7proteinexhibitsonesuchdomainatposition50.HPV-125lackstheregularpRb-bindingcore sequencewithinitsE7protein.InordertoassessthetissuepredilectionandclinicalsignificanceofHPV-125,aquantitative type-specific real-time PCR was developed. The 95% limit-of-detection of the assay was 2.5 copies per reaction (range 1.7–5.7)andtheintra-andinter-assaycoefficientsofvariationwere0.47and2.00for100copiesperreaction,and1.15and 2.15for10copiesperreaction,respectively.TestingofarepresentativecollectionofHPV-associatedmucosalandcutaneous benignandmalignantneoplasmsandhairfollicles(atotalof601samples)showedthatHPV-125isarelativelyrareHPV genotype,withcutaneoustropismetiologicallylinkedwithsporadiccasesofcommonwarts.
