Methods:Osteoblastsfrom6-weekoldSprague-Dawleyratswereculturedontissueculturepolystyrene(TCPS)or ontitanium(Ti)diskswithtwodifferentsurfacetopographies,asmoothpretreated(PT)surfaceandacoarsegrit- blasted/acid-etched(SLA)surface,andtreatedwith1a,25(OH)2D3,E2,orE2conjugatedtobovineserumalbumin (E2-BSA). Results:MaleandfemalecellsrespondedsimilarlytoTimicrostructurewithrespecttocellnumberandlevelsof osteocalcin,transforminggrowthfactor-b1,osteoprotegerinandprostaglandinE2intheirconditionedmedia, exhibitingamoredifferentiatedphenotypeonSLAthanonPTorTCPS.E andE -BSAincreaseddifferentiationand 2 2 localfactorproduction,aneffectthatwasmicrostructuredependentandfoundonlyinfemaleosteoblasts.1a,25 (OH)2D3increasedosteoblastdifferentiationandlocalfactorproductioninfemaleandmalecells,buttheeffect wasmorerobustinmalecells. Conclusions:Maleandfemaleratosteoblastsrespondsimilarlytosurfacemicrostructurebutexhibitsexual dimorphisminsubstrate-dependentresponsestosystemichormones.Oestrogenaffectedonlyfemalecellswhile 1a,25(OH)2D3hadagreatereffectonmalecells.Theseresultssuggestthatsuccessfulosseointegrationinmales andfemalesmaydependontheimplantsurfacedesignandcorrectlevelsofcalciotropichormones.
