Methodology/PrincipalFindings:AhalophilicesterolyticenzymeLipCderivedfromarcheaonHaloarculamarismortuiwas overexpressedfromEscherichiacoliBL21.Thepurifiedenzymeshowedarangeofhydrolyticactivitytowardsthesubstrates of p-nitrophenyl esters with different alkyl chains (n=2216), with the highest activity being observed for p-nitrophenyl acetate,consistentwiththebasiccharacterofanesterase.TheoptimalesteraseactivitieswerefoundtobeatpH9.5and [NaCl]=3.4Mor[KCl]=3.0Mandataround45uC.Interestingly,thehydrolysisactivityshowedaclearreversibilityagainst changes in salt concentration. At the ambient temperature of 22uC, enzyme systems working under the optimal salt concentrationswereverystableagainsttime.Increaseintemperatureincreasedtheactivitybutreduceditsstability.Circular dichroism(CD),dynamiclightscattering(DLS)andsmallangleneutronscattering(SANS)weredeployedtodeterminethe physicalstatesofLipCinsolution.Asthesaltconcentrationincreased,DLSrevealedsubstantialincreaseinaggregatesizes, but CD measurements revealed the maximal retention of the a-helical structure at the salt concentration matching the optimalactivity.TheseobservationsweresupportedbySANSanalysisthatrevealedthehighestproportionofunimersand dimersaroundtheoptimalsaltconcentration,althoughthecoexistentlargeraggregatesshowedatrendofincreasingsize withsaltconcentration,consistentwiththeDLSdata.
