ThereareanumberofinvitrotechniquesforhumanCPepithelium(CPE)andallhavelimitationsthatdonot excludethemapriori,butthatshouldbecarefullytakenintoconsideration.Developmentalandmorphological studieshavebeenlargelyperformedonhumanchoroidplexussamplesofeitherembryonicorpost-mortemorigin. FunctionaluptakestudiesmaybeperformedonpathologicallyunalteredCPsamplesobtainedduringsurgical removalofchoroidplexustumors.Thisapproachcanbeusedtoexploretransportprocessesmainlyacrossthe apicalsideoftheCPE,butcannotbeusedtostudyvectorialtransportacrosstheCPE.Also,thesesampleshave limitedviability.AmonolayerofCPEinculture,grownonpermeablesupports,providesthebestavailabletoolto studytransportprocessesorpolarizedsecretionbytheCP,butthusfaronlylimitedattemptstoculturethesecells havebeenpublishedandtheymainlyincludedatafromneoplasticCPE.Astudythatusedahuman papilloma-derivedcelllineincultureshowedthatitformsamonolayerwithbarrierproperties,althoughthecells expresspleomorphicandneoplasticfeaturesandlackcontactinhibition.OthercellculturesexpresssomeCPE markersbutdonotdeveloptightjunctions/barrierproperties.Thisarticlereviewsthemaincharacteristicsand limitationsofavailableinvitromethodstostudyhumanCPE,whichcouldhelpresearcherschooseanappropriate experimentalapproachforaparticularstudy.
