Huangetal.BMCPlantBiology2012,12:30 /1471-2229/12/30 RESEARCH ARTICLE OpenAccess Dissectinggeneticarchitectureofgrape proanthocyanidincompositionthrough quantitativetraitlocusmapping Yung-FenHuang1,2,AgnèsDoligez1*,AlexandreFournier-Level1,5,Lo?c LeCunff1,3,YvesBertrand1, AurélieCanaguier4,CécileMorel1,ValérieMiralles1,FrédéricVeran2,Jean-MarcSouquet2,VéroniqueCheynier2, NancyTerrier2andPatriceThis1 Abstract Background:Proanthocyanidins(PAs),orcondensedtannins,areflavonoidpolymers,widespreadthroughoutthe plantkingdom,whichprovideprotectionagainstherbivoreswhileconferringorganolepticandnutritivevaluesto plant-derivedfoods,suchaswine.However,thegeneticbasisofqualitativeandquantitativePAcomposition variationisstillpoorlyunderstood.ToelucidatethegeneticarchitectureofthecomplexgrapePAcomposition,we firstcarriedoutquantitativetraitlocus(QTL)analysisona191-individualpseudo-F1progeny.Threecategoriesof PAvariableswereassessed:totalcontent,percentagesofconstitutivesubunitsandcompositeratiovariables.For ninefunctionalcandidategenes,amongwhicheightco-locatedwithQTLs,weperformedassociationanalyses usingadiversitypanelof141grapevinecultivarsinordertoidentifycausalSNPs. Results:MultipleQTLanalysisrevealedatotalof103and43QTLs,respectivelyforseedandskinPAvariables.Loci weremainlyofadditiveeffectwhilesomelociwereprimarilyofdominanteffect.Resultsalsoshowedalarge involvementofpairwiseepistaticinteractionsinshapingPAcomposition.QTLsforPAvariablesinskinandseeds differedinnumber,position,involvementofepistaticinteractionandalleliceffect,thusrevealingdifferentgenetic determinismsforgrapePAcompositioninseedsandskin.AssociationresultswereconsistentwithQTLanalysesin mostcases:fouroutofninetestedcandidategenes(VvLAR1,VvMYBPA2,VvCHI1,VvMYBPA1)showedatleastone significantassociationwithPAvariables,especiallyVvLAR1revealedasofgreatinterestforfurtherfunctional investigation.SomeSNP-phenotypeassociationswereobservedonlyinthediversitypanel. Conclusions:ThisstudypresentsthefirstQTLanalysisongr
