Methods:In this experiment, five differentmethods com- monly used to extract DNA from soilwere chosen to extract DNA from paddy soi.l Then the quality and quantity ofDNA were compared to estimate the fivemethods. Results:DNA extracted by fivemethodswere all greater than 48kb in size, but the yieldswere differen.t Subsequently, they were directly used as tem- plates forPCR amplification without purification, using primers of the bacterial16S rDNA gene. Conclu- sion:Bourrainmethod andMartin-Laurentmethod can generateDNA quicker than othermethods. M artin- Laurentmethod can generate the largest amount ofDNA.
