高效液相色谱法同时测定归芍地黄丸中四种化学成分的含量 黄晓燕1，李玲1，2，李荣玮1，高容1，胡亮1，丁野1基金项目： 基金项目：国家药典委员会《中国药典》2015年版药典科研课题 作者简介：黄晓燕，女，药师，主要从事中药检验及质量标准研究， E-mail: xiaoyanzi_1203@163.com （1. 湖南省食品药品检验研究院，长沙 410001；2. 中南大学药学院，长沙 410013） 摘要：目的 建立高效液相色谱法同时测定归芍地黄丸中莫诺苷、马钱苷、芍药苷和丹皮酚的含量。方法 色谱柱为 Waters Symmetry C18 ( 250 mm ×4.6 mm，5 μm），流动相为乙腈－0.3%磷酸溶液，梯度洗脱，柱温为35 ℃，流速为0.8 mL·min-1，检测波长为240 nm，外标法计算含量。结果 莫诺苷进样量在0.010 4～2.62 μg内与峰面积呈良好的线性关系（r=0.999 9），平均加样回收率为102.8%， RSD为1.4%；马钱苷进样量在0.008 6～2.17 μg内与峰面积呈良好的线性关系（r=0.999 9），平均加样回收率为103.6%，RSD为1.1%；芍药苷进样量在0.008～1.15 μg内与峰面积呈良好的线性关系（r=0.9943），平均加样回收率为102.3%，RSD为2.1%；丹皮酚进样量在0.018～2.25 μg内与峰面积呈良好的线性关系（r=0.999 8），平均加样回收率为102.2%，RSD为1.6%。结论 该方法准确，简便，可行，重复性好，可有效控制归芍地黄丸的质量。 关键词： 归芍地黄丸；高效液相色谱法；莫诺苷；马钱苷；芍药苷；丹皮酚 HPLC simultaneous determination of four chemical components in Guishao Dihuang Pills Abstract: Objective To develop an HPLC method for simultaneous determination of morroniside、loganin、paeoniflorin and paeonol in Guishaodihuang Pills. Methods The column was Waters Symmetry C18 (250 mm×4.6 mm,5 μm), and the mobile Phase was acetonitrile and 0.3% phosphoric acid with gradient elution. The column temperature was 35 ℃, the flow rate was 0.8 mL·min-1, and the detection wavelength was 240 nm, and external standard method was used. Results The method displayed a good linearity at 0.010 4-2.62 μg（r=0.999 9）for morroniside，0.008 6-2.17 μg（r=0.999 9）for loganin，0.008-1.15μg（r=0.994 3）for paeoniflorin and 0.018-2.25 μg（r=0.999 8）for paeonol. The average recoveries were 102.8%(RSD=1.4%),103.6% (RSD=1.1%),102.3%(RSD=2.1%)and 102.2%（RSD=1.1%), respectively. Conclusion This method was accurate and simple, feasible and with good repeatability, and can be used in the quality control of the Guishao Dihuang Pills.
