Abstract: The conditions of purification, denaturation and renaturation of recombinant Bovine grow th hor
mone(BGH) were researched in order toget agood renaturation result. The fermentative thalluseswere disrupted
through highpressure homogenization, and the inclusion body obtainedby centrifugalizationwas denatured in7.
5 mol/L urea for 24 h, dilutedw ith 100 mmolL Tris HCl at pH 10. 0 until the urea concentration was 4. 5
molL, and renatured at4 for48 h. Dialysing 20mmolL Tris HClatpH 9. 0 and removing salt could suc
cessfully renature the recombinantBGH. The renatured recombinantBGH was concentratedw ith 100K and 5 K
ultrafilters and purifiedby fast protein liguid chromatography(FPLC), and thus the targetprotein of high purity
could be got.
