精神分裂症患者富马酸喹硫平血浓度测定及其与疗效的关系 1 2 傅崇铭 郭瑞臣 1 2 山东省精神卫生中心（济南 250014 ） 山东大学齐鲁医院临床药理研究所（济南，250012 ） 摘要 目的：建立反相高效液相色谱法测定精神分裂症患者富马酸喹硫平血浓度的方法学，探讨其血浓度与 患者年龄、性别、服药剂量及临床疗效的关系。方法：采用高效液相色谱法测定 76 例单一服用富马酸喹硫平 的精神分裂症住院患者治疗前及治疗后第2 、4 、6 周肘静脉血浓度，并进行BPRS量表评定及相关性分析。结 果：富马酸喹硫平血浓度在 50~500 ng.ml-1范围内有良好线性关系(r=0.9850) 。在日剂量50~450mg范围内血浓 度随剂量的增大而升高，但与服药患者年龄、性别无显著相关性。血药浓度在 126~350 ng.ml-1范围内临床效 果较好。结论：测定方法简便、准确、专一性强，可用于富马酸喹硫平治疗血浓度监测。治疗窗浓度为 126~350 -1 ng.ml 。 关键词 富马酸喹硫平；血药浓度；高效液相色谱法 A study on serum concentration and clinical response of quetiapine fumarate patients with of schizophrenia 1 Fu Chongming 2 Guo Ruichen 1 Pharmacy of Shandong Mental Health Center(Jinan 250014) 2Dept. of Clinical Pharmacology, Qilu Hospital of Shandong University(Jinan 250012) ABSTRACT: OBJECTIVE A RP-HPLC method was established for the determination of serum quetiapine fumarate concentration in patients with treating schizophrenia ,and to study its relationship with clinical response. METHODS A HPLC method was used to determine quetiapine fumarate serum concentration .76 schizophrenia inpatients before or at the end of 2、4 、6 weeks of quetiapine fumarate treatment and was measured the therapeutic efficacy by BPRS. RESULTS The standard curve was linear within the concentration range of 50~500 ng.ml-1 of quetiapine fumarate in serum (r=0.9850). The serum concentrations are proportionally increased with daily dose in range of 50~450mg, but no -1 significant correlation with age and sex. The optimal range estimated was from 126 to 350 ng.ml .
